Vibrio cholerae possesses multiple quorum-sensing (QS) systems that control virulence and biofilm formation among other traits. At low cell densities, when QS autoinducers are absent, V. cholerae forms biofilms. At high cell densities, when autoinducers have accumulated, biofilm formation is repressed, and dispersal occurs. Here, we focus on the roles of two well-characterized QS autoinducers that function in parallel. One autoinducer, called cholerae autoinducer-1 (CAI-1), is used to measure Vibrio abundance, and the other autoinducer, called autoinducer-2 (AI-2), is widely produced by different bacterial species and presumed to enable V. cholerae to assess the total bacterial cell density of the vicinal community. The two V. cholerae autoinducers funnel information into a shared signal relay pathway. This feature of the QS system architecture has made it difficult to understand how specific information can be extracted from each autoinducer, how the autoinducers might drive distinct output behaviors, and, in turn, how the bacteria use QS to distinguish kin from nonkin in bacterial communities. We develop a live-cell biofilm formation and dispersal assay that allows examination of the individual and combined roles of the two autoinducers in controlling V. cholerae behavior. We show that the QS system works as a coincidence detector in which both autoinducers must be present simultaneously for repression of biofilm formation to occur. Within that context, the CAI-1 QS pathway is activated when only a few V. cholerae cells are present, whereas the AI-2 pathway is activated only at much higher cell density. The consequence of this asymmetry is that exogenous sources of AI-2, but not CAI-1, contribute to satisfying the coincidence detector to repress biofilm formation and promote dispersal. We propose that V. cholerae uses CAI-1 to verify that some of its kin are present before committing to the high-cell–density QS mode, but it is, in fact, the broadly made autoinducer AI-2 that sets the pace of the V. cholerae QS program. This first report of unique roles for the different V. cholerae autoinducers suggests that detection of kin fosters a distinct outcome from detection of nonkin.

Tracking surveys serve as tools to inform programming by routinely collecting data from cross-sections of populations at risk for HIV/AIDS and other adverse health outcomes. This survey among injecting drug users (IDUs) aimed to: 1) identify and describe segments of the population in terms of needle sharing in the three months prior to the survey; 2) understand the needle sharing behaviors of IDUs; 3) explore the sexual behaviors and condom use of IDUs; and 4) apply study findings to interventions designed for reducing the risk of HIV transmission among IDUs. Respondent-Driven Sampling (RDS) was used to recruit 417 male IDUs aged 15 to 45 who live in Uong Bi and Mong Cai towns in Quang Ninh province and Thai Nguyen City in Thai Nguyen province. A screening questionnaire and main questionnaire were the survey instruments. The questionnaire collected information on socio-demographic characteristics, drug use and needle sharing behaviors, opinions about needle-sharing practices, beliefs and knowledge about HIV infection, sexual activity and condom use, and exposure to harm reduction and VCT services. Multivariate analyses were performed to identify significant factors associated with needle sharing and condom use. Simple frequencies and means were run on descriptive data to permit monitoring of project indicators.

The ProstateX dataset (both training and testing cases) have been included in the PI-CAI Public Training and Development dataset. As such, ProstateX as a benchmark has been deprecated and is superseded by the PI-CAI challenge. PI-CAI is an all-new grand challenge, with over 10,000 carefully-curated prostate MRI exams to validate modern AI algorithms and estimate radiologists' performance at clinically significant prostate cancer detection and diagnosis. Key aspects of the study design have been established in conjunction with an international, multi-disciplinary scientific advisory board (16 experts in prostate AI, radiology and urology) - to unify and standardize present-day guidelines, and to ensure meaningful validation of prostate-AI towards clinical translation. The PROSTATEx Challenge ("SPIE-AAPM-NCI Prostate MR Classification Challenge”) focused on quantitative image analysis methods for the diagnostic classification of clinically significant prostate cancers and was held in conjunction with the 2017 SPIE Medical Imaging Symposium. PROSTATEx ran from November 21, 2016 to January 15, 2017, though a "live" version has also been established at https://prostatex.grand-challenge.org which serves as an ongoing way for researchers to benchmark their performance for this task.
The PROSTATEx-2 Challenge ("SPIE-AAPM-NCI Prostate MR Gleason Grade Group Challenge" ) ran from May 15, 2017 to June 23, 2017 and was focused on the development of quantitative multi-parametric MRI biomarkers for the determination of Gleason Grade Group in prostate cancer. It was held in conjunction with the 2017 AAPM Annual Meeting (see http://www.aapm.org/GrandChallenge/PROSTATEx-2).

This collection is a retrospective set of prostate MR studies. All studies included T2-weighted (T2W), proton density-weighted (PD-W), dynamic contrast enhanced (DCE), and diffusion-weighted (DW) imaging. The images were acquired on two different types of Siemens 3T MR scanners, the MAGNETOM Trio and Skyra. T2-weighted images were acquired using a turbo spin echo sequence and had a resolution of around 0.5 mm in plane and a slice thickness of 3.6 mm. The DCE time series was acquired using a 3-D turbo flash gradient echo sequence with a resolution of around 1.5 mm in-plane, a slice thickness of 4 mm and a temporal resolution of 3.5 s. The proton density weighted image was acquired prior to the DCE time series using the same sequence with different echo and repetition times and a different flip angle. Finally, the DWI series were acquired with a single-shot echo planar imaging sequence with a resolution of 2 mm in-plane and 3.6 mm slice thickness and with diffusion-encoding gradients in three directions. Three b-values were acquired (50, 400, and 800), and subsequently, the ADC map was calculated by the scanner software. All images were acquired without an endorectal coil.
