    Nutrient inputs to surface waters are particularly varied in urban areas, due to multiple nutrient sources and complex hydrologic pathways. Because of their close proximity to coastal waters, nutrient delivery from many urban areas can have profound impacts on coastal ecology. Relatively little is known about the temporal and spatial variability in stoichiometry of inorganic nutrients such as dissolved silica, nitrogen, and phosphorus (Si, N, and P) and dissolved organic matter in tropical urban environments. We examined nutrient stoichiometry of both inorganic nutrients and organic matter in an urban watershed in Puerto Rico served by municipal sanitary sewers and compared it to two nearby forested catchments using samples collected weekly from each river for 6 years. Urbanization caused large increases in the concentration and flux of nitrogen and phosphorus (2- to 50-fold), but surprisingly little change in N:P ratio. Concentrations of almost all major ions and dissolved silica were also significantly higher in the urban river than the wildland rivers. Yield of dissolved organic carbon (DOC) was not increased dramatically by urbanization, but the composition of dissolved organic matter shifted toward N-rich material, with a larger increase in dissolved organic nitrogen (DON) than DOC. The molar ratio of DOC:DON was about 40 in rivers draining forested catchments but was only 10 in the urban river. Inclusion of Si in the assessment of urbanization’s impacts reveals a large shift in the stoichiometry (Si:N and Si:P) of nutrient inputs. Because both Si concentrations and watershed exports are high in streams and rivers from many humid tropical catchments with siliceous bedrock, even the large increases in N and P exported from urban catchments result in delivery of Si, N, and P to coastal waters in stoichiometric ratios that are well in excess of the Si requirements of marine diatoms. Our data suggest that dissolved Si, often neglected in watershed biogeochemistry, should be included in studies of urban as well as less developed watersheds due to its potential significance for marine and lacustrine productivity.

    Description

    Previous work has reported that the peptide derived from LfcinB, R-1-R exhibited anti-Candida activity, which is enhanced when combined with an extract from the Bidens pilosa plant. However, the mechanism of action has not been studied until now. In the current study, an approach to the mechanism of action was carried out through the use of mutant strains of C. albicans, a proteomic approach, followed by a bioinformatic analysis, and a validation of the results obtained, through biological assays in both SC5314 strain and a fluconazole-resistant isolate of Candida albicans, after incubation with the treatments. The results obtained with the mutant strains suggest that the R-1-R peptide and the B. pilosa extract exhibit a mechanism of action related to cell wall stress mediated by MAPKs (Hog1p and Mkc1p) and alteration in mitochondrial respiration due to interruption of ETC. After, changes in the protein expression profile of FLC-sensitive and -resistant C. albicans after six h of incubation with R-1-R or combination were analyzed, using a label free quantitative proteomic approach. The proteomic data showed that after treatment with R-1-R in the two study strains, most of the differentially expressed proteins in comparison to controls were up-regulated. These proteins were mainly involved in membrane and cell wall biosynthesis, membrane transporters, oxidative stress, the mitochondrial respiratory chain, and response to DNA damage. Besides, the proteomic analysis of C. albicans parental strain SC5314 treated with R-1-R in combination with an ethanolic extract of B. pilosa was also undertaken. The proteins differentially expressed after this treatment were involved in similar functional processes as the proteins differentially expressed after treatment with R-1-R peptide alone, but at different levels and were mostly down-regulated. The biological assays allowed validation of the proteomic results, evidencing damage to the cell surface, generation of reactive oxygen species and decrease in mitochondrial membrane potential. The results provide information to begin to unlock the understanding of the complex antifungal mode of action of the R-1-R peptide, its combination with the extract, and possibly other derivatives of natural products.

    The R-SCRIPT shows how to extract incubation bouts (i.e., the time between the arrival of a parent and its departure from the nest followed by incubation by its partner) from instantaneous continusou RFID data and from video data scored for start and end of an event on the nest (be it incubation, feeding, exchange, etc.). For this data extraction I recommend, to click always on the bottom line of the red rectangle, as there is always data available due to a dummy variable ("lin") that creates continuous data at the bottom of the active panel. The data are captured only if greenish vertical bar appears and if new line of data appears in R console). # 3 - to extract incubation bouts, first click in the new plot has to be start of incubation, then next click depict end of incubation and the click on the same stop start of the incubation for the other sex. If the end and start of incubation are at different times, the data will be still extracted, but the sex, logger and bird_ID will be wrong. These need to be changed manually in the csv file. Similarly, the first bout for a given plot will be always assigned to male (if no data are present in the csv file) or based on previous data. Hence, whenever a data from a new plot are extracted, at a first mouse click it is worth checking whether the sex, logger and bird_ID information is correct and if not adjust it manually. # 4 - if all information from one day (panel) is extracted, right-click on the plot and choose "stop".
