The dataset includes results of laboratory analyses of 247 wood samples collected without bark from 24 live trees and 52 fallen logs of eight tree species: Acer platanoides, Betula pendula, Fraxinus excelsior, Picea abies, Populus tremula, Quercus robur, Tilia cordata, and Ulmus glabra. The dataset contains the following characteristics of wood samples: (1) bulk density (kg m-3) calculated by dividing the dry mass (at 60ºC for 96 h) by the fresh volume of a sample; (2) pH value measured with an electrode in the aqueous supernatant (0.5 g milled wood sample mixed with demineralized water 1:25); (3) concentrations of C and N (%) determined with a CHN analyser, and (4) concentration of Al, Ca, K, Cu, Mg, Mn, P, and Zn (mg kg-1) defined with ICP-OES. For each wood sample, the stage of decomposition is indicated; it varies from the initial class 0 (living trees) and then from the least decomposed class 1 to the most decomposed class 5. For fallen logs and standing trees, diameter at 1.3 m from the trunk base is given. For logs, the total length and mean stage of decomposition, weighted by the length of longitudinal fragments at different decay stages, are also presented. The logs were formed as a result of a mass windthrow occurred in a mesic broad-leaved forest in the strict Nature Reserve "Kaluzhskie Zaseki" in June 2006. The samples were collected in 2020-2022. Some traditional research methods have the problems of heavy workload and over-reliance on manual work. In view of the above problems, this paper studies the construction of carbon sequestration measurement model based on biomass estimation model. In this paper, sampling design, project boundary, carbon pool selection and other factors are considered in the construction of the model. At the same time, in view of the problem that the calculation results of the actual net carbon sequestration caused by CH4, N2O and other gas emissions in forestry carbon sequestration projects are not accurate enough, this study deduces a specific calculation model based on the actual project development needs and the data of 10 forest farms that have formed forestry carbon tickets. Taking Chinese fir as the research object, the effect of carbon sequestration model was verified and analyzed. Compared with the existing forestry carbon sequestration project evaluation, the results showed that the average relative error of the model was 6. 09%, and the absolute error range was 0. 348-4. 262/hm2, and the model effect was good. The establishment of the model not only solves the problems of over-reliance on manpower, material resources and inefficiency in the development of forestry carbon sequestration in China, but also accelerates the rapid and vigorous growth of forestry carbon sequestration industry in Guizhou, and contributes China's wisdom and effective solutions to global climate governance.

Efforts to characterize microbial life across diverse environments have progressed tremendously, yet the microbiome of Earth’s largest biomass reservoir—the wood of living trees has been largely unexplored. Current understanding of the tree microbiome is largely confined to roots and leaves, with little attention given to the endophytic microbiome of wood. Emergent studies have indicated this zone as a niche for unique taxa, of consequence for ecosystem health and global biogeochemical cycles. The lack of investigation derives partly from the physical recalcitrance of wood, which presents challenges during sampling, homogenization, and the extraction of nucleic acids. In response to these issues, we present an optimized method for processing wood for use in microbial analyses, from sampling through downstream PCR-based analyses. Using methane-cycling taxa as model endophytes, we assess losses in recovery during our method, and determine a limit-of-detection of approximately 500 cells per 100 mg of (dry) wood. For all six species evaluated—which represented genera of broad and needle-leaf angiosperms and gymnosperms—PCR inhibition proved minimal, and we expect this method to be applicable for a majority of tree species. The methods presented herein can facilitate future investigation into the wood microbiome and global microbial ecology of methane cycling.
